Abstract
Background: Mercury has been documented as an industrial risk that posed a serious danger to
human health. Mercury exposure results in oxidative stress that may lead to the pathogenesis of
male reproductive dysfunction. The present study investigated the ameliorating potential
of allium aqueous extract (leek extract). and allium aqueous extract loaded on selenium
nanoparticle against mercuric chloride-induced oxidative deterioration of reproductive functions
in adult male rats.
Methods: Thirty adult rats were divided into five groups: group I (control) received saline.
Group II(HgCl2) received Mercury (15 mg/kg b.w, i.p) dissolved in distilled water. Groups III
(leek) and was given oral gavage of leek extract (250 mg/kg b.w) ,group IV (leek-HgCl2)
received mercury chloride with leek extract (250 mg/kg b.w) (pre-administration).Group V (leek-
HgCl2+ SeNPs) was treated with leek extract loaded on selenium nanoparticle (250 mg/kg b.w)
and mercury chloride. After 14 days of the treatment, the rats were dissected and their testicular
tissue and were used for biochemical analysis while blood plasma was used for protein
determination.hormones (testosterone ,LH and FSH) ,antioxidant markers (SOD,CAT,GSH,GPx
and GR),oxidative stress markers (MDA and NO),inflammatory mediators (TNF-a,IL-1b ,and
NF-kB p65),and apoptosis mediators (BCL-2,BAX and caspases-3 )
Results: The applied dose-treatment of Mercury-induced oxidative stress in the testis and cauda
epididymis tissues of the rats was apparent by a noteworthy decrease in testosterone, LH, FSH,
and PCNA level while there was increase in MDA and NO, GSH, SOD levels also decreased in
CAT, GPX, and GRD level.
The results of the extract treatment group were compared with those of mercury chloride group,
and we figured out that treatment with leek extract loaded on SeNPs significantly ameliorated
testosterone, LH, FSH, and PCNA level Similarly, the level of MDA and NO decreased
significantly, unlike GSH, and SOD levels was significantly improved and the daily.
Furthermore, leek extract loaded on SeNPs administration significantly protected Mercury-
induced testis damage.
Conclusion: Allium porrum(leek) extract showed protection against Mercury-induced oxidative
stress by its antioxidant activity specially when loaded on selenium nano particle. This suggests
that Allium porrum(leek) could be beneficial against toxicity induced by an environmental
toxicant.
